small RNAs were extracted from total RNA and treated with wild-type and D135S AlkB proteins to remove methylation from RNAs. Half of the Alkb-treated small RNAs were saved as input, while the remaining were treated with 0.1M NaBH4 for 30 min on ice in the dark and precipitated with ethanol. The NaBH4-treated RNAs were then treated with an aniline-acetate solution (DEPC H2O: glacial acetate acid: aniline = 7:3:1) at room temperature in the dark for 2 h. The RNAs were purified and employed for cDNA library construction.
