Comprehensive characterization of drug-response, genome, and drug-responsive transcriptome in gastric cancer cell lines

To identify genomic and transcriptomic alterations potentially associated with drug sensitivity and resistance in gastric cancer cells, we performed an integrated phenotypic, genomic, and transcriptomic analysis of a panel of cell lines, including GCIY, GSS, MKN1, IWT1, MKN45, MKN74, and 5-fluorouracil-resistant models, MKN45T and MKN74T. Sensitive and resistant cell lines for each anticancer drug (5-fluorouracil, cisplatin, etoposide, and docetaxel) were determined by 50% growth inhibitory concentration profiling. Cell line-unique driver mutations were identified by targeted panel-based sequencing of 409 cancer-related genes. Drug-responsive genes were identified by RNA sequencing data from these cell lines with or without drug treatments.
