Functional HPV-specific PD-1+ stem-like CD8 T cells in head and neck cancer

The goals of this study were to gain a better understanding of the heterogeneity and transcriptional profile of tumor specific and bulk PD1+ CD8 T cells in HPV primary tumors and metatstatic LNs Overall design: Activated (live CD3+ CD19- CD14- CD16- CD4- CD8+) and double tetramer positive cells from primary tumors and metastatic lymph nodes of HPV+ HNSCC patients and as comparison, bulk PD1+ CD8 T cells from primary tumors and metastatic LNs from 6 patients were FACS-purified. Single-cell suspensions of FACS-purified cells were loaded onto the 10X Genomics Chromium Controller. Library construction was performed using the Chromium Single Cell 5' Library Construction Kit followed by sequencing on a HiSeq3000.
