Saccharomyces cerevisiae cells expressing Hoffman syndrome top2 mutations have defects that give rise to non-homologous end-joining with reduced efficiency. This leads to increased gross chromosomal rearrangements (GCRs) at Cas9-induced double-stranded breaks. Whole genome nanopore sequencing was used to identify the types of GCRs formed.
