The relative and synergistic contributions of genetics and environment to inter-individual immune response variation remain unclear, despite its implications for understanding both evolutionary biology and medicine. We used single cell RNA sequencing (scRNA-seq) to quantify the interactive effects of genotype and environment on immune traits by investigating three inbred mouse sttrain rewilded in an outdoor enclosure and infected with the parasite, Trichuris muris Overall design: Single cell suspensions from the mesenteric lymph node were prepared by mashing the tissues individually through a 70µm cell strainer and washed with RPMI from murine mesenteric lymph nodes. 2000 cells from each individual mouse (Block 1 (n = 51); Block 2 (n = 71)) were labelled with the Ab-Hash Tag Oligonucleoutides (HTO) and analyzed using scRNAseq.
