Study was performed in groups of animals including healthy controls, ischemia reperfusion alone, and administration of drug followed by ischemia reperfusion. Samples from liver and isolated hepatocytes intact and one day after reperfusion were analyzed for gene expression in conjunction with other biological variables. Results indicated substantive alteration after drug intervention in multiple gene ontology groups and cellular events and processes after ischemia reperfusion. Three groups of animals were established: healthy control; ischemia reperfusion alone by temporaly clamping of the portal vein; administration of Etanercept 16-18 hrs before ischemia reperfusion and of nitroglycerin as intravenous infusion before and during ischemia reperfusion. Liver samples were obtained from n=3 per condition. Primary hepatocytes were isolated by two-step collagenase perfusion from additional animals n=3 per condition. One day after ischemia reperfusion, RNA was extracted, pooled for each group and processed for the sequencing on Ion Torrent Proton instrument.
