To analyze the effect of type I IFN on global gene expression in differentiating monocytes, we employed microarray-based gene expression profiling. The MACS-purified mouse BM monocytes were cultured in M-CSF and treated with or without IFN for 60 h. The cells were harvested and isolated RNAs were subjected to microarray analyses. MACS-purified monocytes from mouse bone marrow were divided into two groups (each in triplicates) and treated with 20 ng/mL M-CSF, together with or without 100 U of mouse IFN-beta for 60 h. The resulting cultures were harvested and the RNA samples were subjected to microarray analyses.
