金花茶是山茶属内金黄色花朵的一些种类, 为珍稀的种质资源。该研究在金花茶原产地生态调查的基础上, 模拟金花茶生态环境, 在南宁新竹苗圃和南宁树木园各建一基因库, 共占地27亩, 栽母树 1027株, 搜集保存了22个种和变种, 为迄今我国金花茶资源搜集最齐全、管理和生长、开花最好的两座金花茶基因库。该研究采用精密扫描电镜对金花茶叶片、花粉扫描, 发现了各种金花茶花粉的特征差异, 为金花茶科学分类、鉴定提供了微观依据。对金花茶胚胎发育进程的研究发现, 在胫囊发育期及合子分裂后, 有储存淀粉消耗高峰期, 说明了早期落果的原因, 提出了减少落果的措施。又查明卵细胞受精后, 有长达120天的休眠期, 指出组培须在受精4个月后进行的内在根据, 避免了幼胚离体培养取材的盲目性。此外, 筛选了壮苗壮根一次成苗培养基, 缩短了组胚时间。

Camellia species with golden-yellow flowers within the genus Camellia are rare and precious germplasm resources. Based on ecological surveys conducted in the native habitats of Camellia chrysantha, this study simulated its natural ecological environment, and established two germplasm banks respectively at Xinzhu Nursery and Nanning Arboretum in Nanning. The two germplasm banks cover a total area of 27 mu (approximately 1.8 hectares), with 1,027 maternal trees planted, and have collected and preserved 22 species and varieties. These are currently the two most comprehensive and well-performing germplasm banks for Camellia chrysantha resources in China, excelling in resource collection completeness, management, tree growth and flowering. This study employed high-precision scanning electron microscopy (SEM) to scan the leaves and pollen of Camellia chrysantha, and identified distinct characteristic differences among the pollen of different Camellia chrysantha taxa, providing microscopic evidence for the scientific classification and identification of Camellia chrysantha. Research on the embryonic development process of Camellia chrysantha revealed a peak period of stored starch consumption during the embryo sac development stage and after zygote division, which clarified the cause of early fruit drop and proposed corresponding measures to reduce fruit drop. It was also confirmed that after fertilization of the egg cell, there is a dormancy period lasting up to 120 days. This finding established the internal basis for conducting tissue culture 4 months after fertilization, thus eliminating the blindness of sampling for in vitro culture of immature embryos. In addition, a medium for promoting strong seedlings and robust roots that enables one-step seedling formation was screened, shortening the time required for embryo tissue culture.
