Relationship between donor cell chromatin openness and cloned embryo development

The development rate of cloned blastocysts varies significantly among individual donor cells, currently, studies have focused on the status, type, and epigenetic modification status of donor cell. However, differences in SWI/SNF gene expression and chromatin openness in individual cells have not been reported. In this study, we found that donor cells with high blastocyst development rate had higher SWI/SNF family genes expression, ATAC-seq assay revealed more open chromatin, RNA-seq revealed higher expression of genes related to ATPase, combined with the fact that the SWI/SNF complex regulated chromatin structure by utilizing the energy generated by ATP hydrolysis, we finally found that ATP content could affect the expression of SWI/SNF family genes to some extent. The results indicate that the SWI/SNF complex contributed to chromatin opening, and chromatin opening in donor cells was essential for cloned blastocyst development.
