H3K4me3 deposited by the Set1/COMPASS is one of the most studied modifications because of its relevance to active gene expression. Whereas most studies focus on transcriptional activation of H3K4me3, little is known about how the level of H3K4me3 is maintained at the optimal level. We identified a transcriptional regulator Tup1 interacts with FgSet1. To elucidate the molecular mechanism of FgSet1-Tup1 interactions, we performed transcriptome analysis, CUT&Tag-seq and ATAC-seq. Our findings reveal Tup1 acts as a molecular brake precisely preserving homeostasis of transcripts and achieve optimal H3K4me3-marked gene expression.
