This dataset represents both clonal phage genome and wild virus metagenome data used to validate Linker amplification for quantitative metagenomics of wild viruses and other ultra-low concentration DNA samples. The project was an assessment of a DNA amplification method intended for metagenomic samples prior to next-gen sequencing. We tested the method by sequencing a single genome isolate (Pseudoalteromonas H105/1) and a single wild virus community/metagenome sample (Biosphere2 ocean). For each, the isolate and the virus community, we tested several parameters by varying amplification cycle numbers and a PCR reconditioning step. Marine phage was isolated from North Sea seawater at 1m depth, off the coast of Helgoland, Germany (54.17 N, 7.88 E); host Pseudoalteromonas sp. H105; collected Sep-1990, kindly provided by A. Wichels (AWI Helgoland). For the isolate, there are 30 treatments of the same original phage isolate plus 1 unamplified (31 data sets total) For the virus community sample, there are 8 treatments of the same original water sample plus 2 unamplified (10 data sets total).
