We established a de novo approach of generating T cells from mouse ESC/iPSC. mRNA profiles of T cell receptors (TCR), including TCR of induced CD4SP/CD8SP sorted from spleen or thymus. 15000 of each cell type were sorted for RNAseq analysis according to kit instruction of SMARTer Mouse TCR a/b Profiling Kit (Clontech) , using Illumina MiSeq.
