T lymphocytes are required for adaptive immune responses and play a pivotal role in immune surveillance. However, the characteristics of T-cell receptors (TCR) remain largely unexplored in granulomatous lobular mastitis (GLM). Consequently, evidence for local and peripheral immune status in GLM is also lacking. Here we investigated the TCR repertoire using high-throughput sequencing in nipple discharge or peripheral blood collected from 97 GLM patients and 24 mammary gland hyperplasia (MGH) as control. As compared to peripheral blood, TCR repertoire in nipple discharge is dominated by a small number of clonotypes, which leads to significantly lower TCR diversity (P <0.001). Additionally, a series of V and J gene segments were highlighted for significantly differential expression between nipple discharge (ND) and peripheral blood (Blood). This study demonstrated the unique property of TCR repertoire in peripheral blood, which provided possibility for future improvements in immune-related diagnosis and therapy for GLM.
