Quercus fabri is a common timber oak tree species widely distributed in subtropical areas of China. In this study, we present a chromosome-scale reference genome assembly of Q. fabri achieved by integrating PacBio Sequel II, DNBseqTM and Hi-C sequencing platforms. Our findings indicated that the PacBio draft genome has a size of 836.74 Mb, with a BUSCO complete gene percentage of 98.8%. Hi-C scaffolding successfully anchored 816.69 Mb of contigs (97.6% of the draft assembly) onto 12 pseudochromosomes. The contig N50 and scaffold N50 values were 61.8 Mb and 69.2 Mb, respectively. A total of 37,182 protein-coding genes were predicted and annotated.
