We performed single-cell RNA sequencing (scRNA-seq) to dissect the heterogeneity of lung cells from mice exposed to papain, with or without co-administration of TLR ligands or bacterial extracts, identify key transcriptional signatures associated with type 2 inflammation, and uncover molecular changes induced by cross-regulatory signals. Overall design: Experimental design : To assess acute effects, long-term changes, and recall responses induced by allergen exposure and cross-regulation, we performed scRNA-seq on whole lung cells from mice under six conditions: 1.Acute Phase (Day 3): Papain or papain+CpG, analyzed 3 days after administration. 2.Long-Term Phase (6 Weeks): Papain or papain+CpG, analyzed 6 weeks post-administration (no recall). 3.Long-Term Phase with Recall: Papain or papain+CpG, followed by a secondary papain challenge at 6 weeks.
