This study was designed to investigate the potential immunomodulatory properties of a novel pleuromutilin derivative, APTM (14-O-[(4-amino-6-hydroxy-pyrimidine-2-yl) thioacetyl] mutilin), beyond its established antibacterial activity. To explore this, we established an *in vitro* model of inflammation by stimulating the murine macrophage cell line RAW 264.7 with lipopolysaccharide (LPS) and subsequently performed transcriptome-wide analysis using RNA sequencing (RNA-Seq) to profile gene expression changes following APTM treatment. The primary objective of this project is to identify the molecular pathways and key genes regulated by APTM during an inflammatory response. The resulting dataset offers a comprehensive resource for understanding the anti-inflammatory mechanism of APTM, providing a foundation for its potential development as a therapeutic agent with dual antibacterial and anti-inflammatory functions.
