Plant cell walls are highly dynamic structures, playing a critical role in diverse biological processes, including cell morphogenesis and plant-pathogen interactions. During these processes, plant cell wall integrity (CWI) can be impaired. Thus, it is important for plants to consistently monitor the state of their cell walls and ensure their functional integrity. Here, cellulose biosynthesis inhibition triggered by isoxaben (ISX) was employed to investigate the transcriptional reprograming upon cell wall damage (CWD) in Arabidopsis thaliana seedlings. Analysis of mutants impaired in CWI indicates that MALE DISCOVERER 1-INTERACTING RECEPTOR LIKE KINASE 2 (MIK2) is a key regulator of ISX-induced transcriptional reprograming. RNA-seq-based transcriptomics showed that disruption of CWI promotes pattern-triggered immunity (PTI) through transcriptional reprograming mediated by the receptor kinase MIK2 in Arabidopsis.
