LINE1 and PRC2 control nucleolar organization and repression of the 8C-state in human ESCs [RNA-Seq_RSeT+DT H9_EZH2i]

In this study, we investigated PRC2 (polycomb repressive complex 2), the H3K27me3 modifier, in naive hES cells for its roles in regulating 8-cell (8C) stage genes expression. EZH2 inhibitor, UNC1999 and its inactive analog UNC2400 were used for the treatment. Naive H9 hES cells were maintained RSeT. 24 hours after cell splitting, cells were maintained in the RSeT media added with 10 nM DZNep and 5nM TSA (RSeT+DT), in combination with UNC1999 and UNC2400 added at 2.5 μM final concentration for the treatment and controls. 48 hours later, cells were directly processed for RNA extraction and RNA-seq library prep.
