This study aimed to investigate the transcriptional responses of human THP-1 monocytes and RPMI-8226 multiple myeloma cells to various innate immune agonists. Cells were treated with ALPK1 agonist (ADP-Hep or UDSP-Hep), TLR7/8 agonist (R848), or STING agonist (ADU-S100) at different concentrations for 4 hours, and RNA-seq was performed to characterise the downstream gene expression programmes triggered by these stimuli. Overall design: THP-1 and RPMI-8226 cells were cultured under standard conditions and exposed to different concentrations of the innate immune agonists ADP-Hep, UDSP-Hep, R848, or ADU-S100 for 4 hours. Total RNA was extracted from treated and untreated control samples, and RNA sequencing was performed using an Illumina platform. This design enables comparison of the gene expression profiles induced by different innate immune pathways across two human cell lines.
