Pharmaceutically applicable polyols are represented with different chemical compounds, with different structures and solid-state properties as well as water content. Water content is an important measured parameter since it can affect chemical stability, quality and safety of the formulation during both manufacturing process and storage.
Method. The water content of polyols was determined by Karl Fischer titration. The accurately weighed samples of approx. 150 mg to approx. 9000 mg (mean 250 mg, depending on the sensitivity of the titrator towards the specific polyol) were analysed using volumetric Karl-Fischer titrator (V10S; Mettler Toledo AG, Switzerland). HYDRANALTM Methanol dry (Honeywell Fluka, Seelze, Germany) and HYDRANALTM Composite 5 (Honeywell Fluka, Seelze, Germany) were used as reagents for volumetric one-component KF titration. Measurements were done in triplicate, and the water content (%, w/w) were presented as averages and standard deviations (n = 3; Av. ± S.D.).
Data. The data for individual polyols in the form of an CSV spreadsheet (file name: KF titration of polyols) was composed of the 3 individual measurements of water content, average water content, the standard deviation (S.D.) and the relative standard deviation (R.S.D.). To simplify the reusing of data, average water content values with S.D. were presented in a graphical format (file name: Figure.jpg).
