Effect of IAA treatment on gene expression and metabolic profiles. Recent reports showed that exogenous application of IAA could regulate the levels of endogenous carotenoids, ABA and IAA in rice27 and colour and texture in peach28. However, there are few reports on the effects of IAA on citrus metabolism. To determine the effect of IAA on citrus metabolism, we treated citrus callus with 100 µM IAA. After treating RM citrus callus with IAA, the expression of 23 genes involved in carotenoids, glycolytic and organic acids metabolism was up-regulated by more than 1.5-fold , and the expression of 18 genes involved in soluble sugar synthesis, d-limonene and phenylpropanoids metabolism were down-regulated by more than 1.5-fold . In addition, the contents of carotenoids and organic acids increased, and the contents of lignins, d-limonene, and soluble sugars decreased, which was consistent with the changes in gene expression levels . Taken together, these results indicated that IAA regulated the genes of several metabolic pathways at the transcriptional level, which in turn led to profile changes in the above fruit quality-related metabolites in the treated callus.
