Toxoplasma gondii Transcriptome and RIP-Seq

We have identified four mtAP2 proteins that localize to the mitochondrial matrix in Toxoplasma gondii. Considering the exclusive nature of AP2 proteins as transcription factors, our study aimed to investigate their involvement in mitochondrial genome transcription. To achieve this, we conducted RNA-seq analyses to evaluate the impact of depleting the four mtAP2 proteins on both mitochondrial and genome-wide transcriptomes. To further identify the constituents of mitochondrial ribosomal RNAs (rRNAs), we employed a technique known as RIP-Seq, which stands for RNA Immunoprecipitation followed by Sequencing. This approach enabled us to identify and analyze the specific rRNA molecules associated with the LSU or SSU.
