Role of IFITM3 during primary influenza infection and reinfection

WT or IFITM3-KO mice were infected with PR8 H1N1 influenza virus and reinfected with X31 H3N2 influenza virus Overall design: Mice were challenged sublethally with 0.15LD50 of PR8 H1N1 virus provided by Carles Martinez, which corresponds to 5 plaque forming units (PFU) given intranasally in 50 microliter volume. This is 10^6 dilution of the original stock. Control mice were mock-challenged with diluted allantoic fluid. Four weeks after PR8 infection, mice were reinfected with 1000PFU of X31 H3N2 virus. Lungs were collected, divided in 5 parts and processed at 3 days post infection (DPI) with PR8 H1N1 virus and at 3DPI and 5DPI with X31 H3N2 virus according to the recommendations of the different groups that will process the sample (virus titration, proteomics, lipidomics,metabolomics, transcriptomics).
