FOXS1 is Increased in Liver Fibrosis and Regulates TGFbeta Responsiveness and Proliferation Pathways in Human Hepatic Stellate Cells

LX2 (human hepatic stellate cells) FOXS1 KO and scrambled control cells were plated in 10% FBS 1%AA DMEM and allowed to grow to 80% confluency. Then, cells were treated with 5ng/mL TGFb or vehicle (10mM Citric acid) for 24 hours. Total RNA was harvested using the RNeasy kit (Qiagen). We quantitated the total RNA on a NanoDrop spectrophotometer (Thermo Fisher Scientific, Wilmington, DE), and samples were shipped on dry ice to Novogene (Sacramento, CA) for mRNA sequencing.
