Most of the endometrial carcinoma (EMC) carry multiple mutations (>30 mutations) even in one tumor, but it is unclear which and how the synergies of mutations contribute to the pathology of the EMC. Conventional EMC mouse models based on Cre-loxP system are costly, and crossing of these lines is extremely laborious and time-consuming. To simplify the introduction of multiple mutations with a high flexibility, we applied in vivo genome editing to the murine endometrium and found that it enables to induce aggressive endometrial tumors within 3 months. We believe that our in vivo approach provides high-throughput in vivo screening for oncogenic potentials of mutations in human cancer and will facilitate new drug discoveries targeting specific molecules in EMCs
