Single cell RNA-seq of CDH5+; Rela fl/fl murine hepatic CD4+ T-cells

Single-cell sequencing of CD4+ T-cells from mouse livers containing RAS-senescent hepatocytes, with or without endothelial-specific knockout of Rela. Overall design: multiplexed single cell RNA-seq experiment: Eight Cdh5:Cre-ERT2;Rela fl / fl mice were used. All eight received a hydrodynamic tail-vein (HDTV) injection of NRAS^G12V-containing transposons, leading to mosaic RAS-senescent hepatocytes. Subsequently, 4 were injected with vehicle control and 4 injected with tamoxifen, leading to endothelial-specific deletion of Rela. CD4+ T cells were flow-sorted from the harvested livers at day 9 post-HDTV injection, labelled with Hashtag oligos, before multiplexing and single cell RNA sequencing.
