Innate immune activation is regulated by cytokines and serves as a protective mechanism, its orchestration and timely resolution is essential for patient recovery. In order to investigate the role of IL-18 in a relevant human model, we cocultivated healthy donor monocytes with human iPSC -derived intestinal organoids and stimulated them with IL-18. Single-cell RNAseq data revealed a prominent effect of the stimulations on organoid tissue remodeling. In order to investigate the effect of IL-18 on monocyte mobility and differentiation we co-cultivated human primary monocytes with 3D human intestinal organoids (IOs) and stimulated them with IL-18. Differential gene expression analysis from single-cell RNAseq data from (IOs) stimulated with IL-18.
