Single-molecule measurements of (i) an optically trapped bead, (ii) the DNA hairpin 30R50/T4, and (iii) a DNA reference construct lacking the hairpin, all measured with optical tweezers. All data files are formatted as space-delimited text, containing columns or rows of data representing the position of the bead along the reaction axis or the extension of the DNA molecule (all values in nm). Each folder contains data from two types of measurement: (a) files ending in "SampleTrajectory.txt", which contain single columns of data representing a segment of the full trajectory, and (b) files ending in "Fluctuations.txt" or "TransitionPaths.txt," which contain segments of the trajectories corresponding to nonproductive fluctuations or transition paths, respectively (individual fluctuations/transition paths are listed in space-delimited rows). All data were sampled at 1 MHz. For the DNA hairpin and the reference construct, the measurement modality (constant force or high stiffness) is listed in the folder containing the data, and also noted in the filename (as "CF" or "HS", respectively). For the DNA hairpin and the reference construct, the data for each molecule is found within a subfolder containing the molecule number (e.g. "Molecule 1") and an identifier is also included in the filename (e.g. "Mol1"). Additional data are available from the authors if needed.
