A 24-Marker Flow Cytometry Panel for Comprehensive Immune Profiling Using Spectral Flow Cytometry

To comprehensively immunophenotype the common and rare peripheral blood mononuclear cell subsets in patients with solid and hematological malignancies treated with immunotherapy Conclusion: This 24 marker panel enables reliable identification of myeloid and monocyte derived suppressor cells (MDSC), basophils, myeloid and plasmacytoid dendritic cells, classical, intermediate and non-classical monocytes, NK cell subsets, CD4 and CD8 conventional T cells, TCRαβ and TCRγδ T cells, Regulatory T cells, B cells and plasma cells from mononuclear cells. Daily instrument QC using SpectroFlo® QC beads prior to acquiring reference controls and samples to ensure optimal performance of the cytometer. Titration of all the markers using the same population, single stained reference controls using beads/cells for unmixing and generating compensation matrix, NxN plots using the controls to assess the accuracy of spill over correction and compensation.
