S. venezuelae plate growth assay 2025

To investigate the phenotypic characteristics, the growth rate and the timing of sporulation onset in the S. venezuelae hupSΔCTD::apra mutants, a plate growth assay was performed. Spore samples of the four mutants FH04, FH08, FH10 and FH11 as well as the S. venezuelae WT and ∆hupS were diluted to the same initial OD of 0,05-0,06 and inoculated onto separate sectors of a MYM agar plate without antibiotics. The plate was incubated at 30°C for 52,5 hours in a sealed growth chamber. To monitor bacterial growth, the plate was automatically imaged every 30 minutes. The images starting from 19,5 hours up to 52,5 hours were compiled to generate this time-lapse video.
