CD52 is a 12 amino acid glycoprotein that is widely expressed on lymphocytes. In T cells, the soluble form of CD52 has been shown to inhibit TCR function by binding Siglec-10. In Monocytes, soluble CD52 has been shown to inhibit TLR signaling and induces apoptosis at high concentrations. To assess the impact of CD52 on B cell function, we left B cells either untreated or exposed to recombinant CD52-Fc for 6 hours prior to lysis and bulk RNA-sequencing. Gene expression analysis revealed that soluble CD52 induced downregulation of immunoglobulin and B cell receptor related genes such as CD19 and CD22. Therefore, soluble CD52 appears to have a suppressive role on B cell function. B cells untreated (n = 6) or CD52-Fc treated (n = 6) for 6 hours at 37C.
