Experimental in vivo hyperpolarization of E14.5 apical progenitors shifted their transcriptional programs with precocious generation of intermediate progenitors, and a forward shift in the laminar, molecular, morphological and circuit features of their neuronal progeny. In particular, we reported an overall repression of L4-type neuronal genes and a premature induction of L2/3-type neuronal genes following Kir2.1 overexpression. Mice were electroporated with pCAG-Kir2.1- IRES-GFP at E14.5 or with pCAG-IRES-GFP at E15.5.FlashTag was injected in these pups after 12h to compare isochronic cohorts of labeled cells within single animals (see Telley et al.,DOI: 10.1126/science.aad8361).
