In this study, we define ZBTB7A as a driver of cellular survival during HCoV-229E infection. Overall design: Huh7-dCas9 cells were expanded and 3Ã10^7 Huh7-dCas9 cells were transduced with the CRISPR activation sgRNA library (Calabrese set A, Addgene#92379) in two replicates. After 1 µg/mL puromycin selection, half of the cells were collected as control samples and the other half of cells were infected with HCoV-229E (MOI=0.02). Surviving cells were then harvested and the genomic DNA in control and surviving cells were extracted. The sgRNA sequences were then amplified and sequenced.
