Single cell RNA-sequencing reveals molecular signatures that distinguish allergic from irritant contact dermatitis

Allergic and irritant contact dermatitis can be challenging to distinguish. We used single cell RNA sequencing (scRNA-seq) and insterstitial fluid proteomic analysis to compare suction blister biopsy skin samples from human volunteers with induced allergic and irritant contact dermatitis reactions. Volunteers were first sensitized to the allergen, squaric acid dibutyl ester (SADBE). Skin reactions were induced by 2-day application of individual patches containing SADBE, an acetone vehicle control, and the irritant sodium lauryl sulfate (SLS). Skin reactions as well as a nonlesional control site were sampled by suction blister biopsy. A 4-day allergic reaction was also sampled by 2-day allergen patch application followed by 2 days of rest before the suction blister biopsy. Suction blister samples were collected by aspirating fluid from the blister. Samples were centrifuged to pellet cells, which were analyzed by scRNA-seq.
