资源简介:
To test the functional impact of Pol III on controlling alternative CPA site selection, we depleted, Rna15p, a component of the cleavage and polyadenylation factor I, Bdp1p, a core component of TFIIIB, and Bdp1p/Rna15p together, from the nucleus and assessed the effect on 3´ UTR lengths of genes nearby tRNAs. Many genes show changes in 3' ends after Bdp1p depletion indicating that transcripts can transcription termination can be due to TFIIIB roadblocks rather than transcript cleavage by cleavage and polyadenylation factors. Overall design: Oxford Nanopore Direct RNA sequencing performed, in triplicate, on wild-type, Bdp1, and Bdp1 Rna15 anchor away strains.
