Tie1 and Tie2 receptors are important in the maintenance of the vascular steady-state. We used scRNAseq to analyze how genetic deletion of Tie1 and Tie2 affects expression of proliferation-related genes in mouse endothelial cells. Overall design: (1) Stromal-vascular fraction (SVF) of the mouse heart was prepared by enzymatic (mix of collagenases) digestion of the heart tissue. RBC were lyzed by RBC-lysis buffer. (2) EC-enriched fraction of the mouse heart was prepared by enzymatic digestion of the heart tissue. EC fraction was isolated using FACS and antibody staining of the EC markers. (3) EC-enriched fraction of the mouse kidney was prepared by enzymatic digestion of the kidney. EC fraction was isolated using FACS and antibody staining of the EC markers. (4) EC-enriched fraction of the mouse lung was prepared by enzymatic digestion of the lung. EC fraction was isolated using FACS and antibody staining of the EC markers.
