In this research, we found mcm5 mutation lead to T cell deficiency and apoptosis in zebrafish. To reveal the molecular mechanism of mcm5 mutation in regulating T cell maturation, we performed RNA-Seq to discover the signal pathways regulated by mcm5. Overall design: In order to reveal the downstream signals of mcm5 regulated in zebrafish, we performed transcriptome analysis of mcm5 mutants and wild type zebrafish. About 150 zebrafishes with 3.5dpf in each group were dived into three replicates, and then total RNA of wild type or mcm5 mutated zebrafish was extracted, and the cDNA library was constructed for sequencing. All the DNA libraries were sequenced by HiSeq X Ten. And the clean data were generated from the original raw data by removing the adapters.
