Analysis of bone-marrow derived macrophages (BMDMs) stimulated with single or combinatorial TLR agonist and MEK1/2 inhibitor in wild-type and Irf1-/- macrophages. Results provide insights into the versatile but yet uncharacterized roles of IRF1 in TLR-MEK1/2-ERK MAPK signaling. BMDMs from wild-type or Irf1-/- C57BL/6J mice were stimulated with TLR7 agonist R848 alone or in combination with MEK1/2 inhibitor U0126 for six hours. Non-treated samples and DMSO were used as controls for R848 and MEK1/2 inhibitor, respectively. Total RNA were extracted and subjected to Clariom™ D assays (previously known as GeneChip™ Mouse Transcriptome Array 1.0).
