Next-generation sequencing analysis of Abcg2-expressing vascular endothelial stem cells and mature endothelial cells

We have identified that ATP binding Cassette Subfamily G Member 2 (Abcg2) lables vascular endothelial stem cells (VESC) that can for endothelial cell (EC) colonies in vitro and functional vessels in vivo. Here we compared the transcriptome between Abcg2-expressing VESC and Abcg2- mature EC from multiple murine tissues. Neonatal or 6-week-old Abcg2CreERT;ROSATdTomato mice were given tamoxifen injection one day before dissection. CD31+CD45-Ter119-TdTomato+ VESC and CD31+CD45-Ter119- mature EC from lung, heart and muscle were sorted by flow cytometry. RNA isolation and next-generation sequencing were then performed.
