The transcriptome of Verticillium dahliae JR2 wild-type cultured in xylem sap was compared versus pectin-rich medium. The JR2 wild-type was pre-cultured shaking at 120 rpm in 50 ml of liquid simulated xylem medium (SXM, modified from Neumann and Dobinson (2003) as described by Hollensteiner et al. (2017)) at 25 C for 5 days. Mycelia were then transferred to either pectin-rich SXM or to extracted tomato xylem sap and further incubated for 8 hours.
