The Parkinsonism-associated protein DJ-1/Park7 has three bacterial homologs Hsp31, YhbO and YajL in Escherichia coli that are encoded by hchA, yhbO and yajL, respectively. A hchA yhbO yajL triple mutant derivative (AOL) of the BW25113 (WT) strain was constructed. To identify mutations of the AOL strains, the genomic DNA sequences of BW25113 and AOL were determined. AOL and WT were grown to stationary phase and genomic DNAs were extracted. Standard paired-end (PE) genomic library were constructed and sequenced on an Illumina HiSeq 4000 Illumina sequencer (PE 125 nt) by GATC-Biotech according to manufacturer protocols.
