Saccharomyces cerevisiae Transcriptome or Gene expression

S. cerevisiae wild-type strain and YMR253C knockout yeasts were inoculated in YPD liquid medium respectively, and cultured on a constant temperature shaker at 200 r/min for 72 h. Three replicate samples were set for wild-type and YMR253C knockout yeast. Afte the yeast was collected by centrifugation, total RNA was extracted and the integrity of RNA was checked by Agilent 2100 bioanalyzer. Then the library was constructedand preliminary qualified using Qubit2.0 Fluorometer. Agilent 2100 bioanalyzer was used to detect the insert size of the library. After the library is qualified, the different libraries are pooled according to the effective concentration and target data volume requirements for Illumina sequencing.
