Melanoma plasticity is regulated by mTOR-mediated crosstalk between MITF and TFE3

Bulk RNAseq was performed to identify differences in gene expression in SKMEL28 cells as related to MITF and TFE3 expression. TFE3KO cell lines were created in the D2-MITFKO SKMEL28 and A375 background via CRISPR-cas9 strategies 500k SKMEL28: WT, D2 MITFKO, D2 MITFKO/TFE3KO (clone 3 and clone 13) and A375: WT and TFE3KO cells were plated in triplicate for biologic replicates. RNA was extracted utilizing RNeasy Plus Kit (Qiagen).
