Weight-Response Trajectories across Cardiovascular and Hepatic Outcomes in GLP-1 Receptor Agonist Trials

This repository contains the trial-level dataset and analytical code supporting a systematic review and meta-regression of cardiovascular outcome trials (CVOTs) evaluating glucagon-like peptide-1 receptor agonists (GLP-1RAs) and related incretin-based therapies. The study examined whether greater achieved weight loss is associated with proportionally greater reduction in major adverse cardiovascular events (MACE), and whether cardiovascular benefit plateaus beyond modest weight loss.Using published data from large, randomized CVOTs reporting adjudicated MACE outcomes, trial-level meta-regression analyses were conducted to explore linear, categorical, threshold-based, and non-linear relationships between between-group weight loss and cardiovascular outcomes. Prespecified sensitivity analyses evaluated the robustness of findings across trial populations and comparator designs.The repository includes the derived trial-level dataset, fully annotated R scripts used for the analyses, and the complete electronic search strategies. These materials are provided to support transparency, reproducibility, and independent validation of the findings.
