The title of the paper is the following: "How different are 2D and 3D in vitro model for investigating the mechanism of action of bioactive compounds? The case of phytocannabinoid CBDA". The scope of the present work concerned the investigation of the CBDA-EIF2A interaction in 3D-U87MG cell model. For pulsedSILAC, U87MG cells were cultured in 3D-cell model in presence or not of CBDA. Subsequently, protein mixture was obtained and click chemistry reaction was carried out. Then, enzymatic digestion with trypsin and peptide clean-up were performed and the samples underwent to LC-MS/MS analysis. Raw files obtained from Orbitrap fusion mass spectrometer were processed with Proteome Discoverer software. Fasta file UP000005640_9606 was set as protein database. The maximum number of missed cleavages was set to 2. Unique peptides were set for quantification. Quantitative analysis was set as ratio Heavy/Medium.
