Early fate of exogenous promoters in E. coli

The study evaluated the impact random insertions of a reporter gene on gene expression and early fate of exogenous promoters in E.coli. Bacterial populations sorted based on GFP fluorescence from FACS experiments were grown in LB media containing the antibiotic Kanamycin for 16 hours. The genomic DNA samples were isolated from these populations using the QIAGEN DNA purification kit protocol. TRADIS sequencing was performed using a 50-cycle single-end MiSeq reagent kit (Illumina).
