收集胃癌患者的血清与尿液,采用测序技术,从血清中筛选出21个差异表达的miRNA,从尿液外泌体中筛选出11个差异表达的miRNA。从胃癌患者血液与尿液外泌体中筛选出11miRNA与早期胃癌发生密切相关。开发了用于早期胃癌筛查的血液或尿液外泌体中10个miRNA标志物同步检测PCR试剂盒。建立了MiR-19b-3p 与MiR-16-5p,miR-92a-3p与miRNA-23b定量PCR检测方法及试剂盒,临床验证表明这4个标志物的表达水平与胃癌密切相关,可辅助胃癌早期筛查。筛选出胃癌血清与尿液外泌体中miRNA标志物,建立了基于量子点编码微球结合流式细胞仪的多个胃癌相关microRNA同步检测的试剂盒,获得CE认证;开发了基于量子点编码微球的七种肿瘤标志物定量分析试剂盒,,获得CE认证,可辅助胃癌早期筛查。开发了胃癌耐药与预后预测的5个miRNA定量PCR试剂盒,可以预测胃癌患者是否存在顺铂、阿霉素、紫杉醇耐药;是否存在复发预后风险。
Serum and urine samples from gastric cancer patients were collected, and sequencing technology was adopted. Twenty-one differentially expressed miRNAs were screened from serum, while 11 differentially expressed miRNAs were identified from urinary exosomes. Eleven miRNAs screened from the blood and urinary exosomes of gastric cancer patients are closely associated with early gastric carcinogenesis. A PCR kit for simultaneous detection of 10 miRNA markers in blood or urinary exosomes was developed for early gastric cancer screening. Quantitative PCR (qPCR) detection methods and kits for miR-19b-3p, miR-16-5p, miR-92a-3p and miRNA-23b were established. Clinical validation demonstrated that the expression levels of these four markers are closely correlated with gastric cancer, and they can assist in early gastric cancer screening. MiRNA markers in serum and urinary exosomes of gastric cancer patients were screened, and a kit for simultaneous detection of multiple gastric cancer-related microRNAs based on quantum dot-encoded microspheres combined with flow cytometry was developed and granted CE certification. A quantitative analysis kit for seven tumor markers based on quantum dot-encoded microspheres was developed, granted CE certification, and can assist in early gastric cancer screening. Five miRNA quantitative PCR kits for gastric cancer drug resistance and prognosis prediction were developed, which can predict whether gastric cancer patients develop resistance to cisplatin, doxorubicin and paclitaxel, as well as the risk of recurrence and prognosis.
