QBC-CyTOF cross-species PBMC dataset: human and rhesus macaque single-cell proteomics datasets
收藏资源简介:
Single-cell protein-profiling dataset accompanying the manuscript 'Split-Pool Synthesis Enables High-Dimensional Cross-Species Immune Cell Profiling and Signaling Quantification at Single-Cell Resolution'. Peripheral blood mononuclear cells (PBMCs) from 3 human donors (H1-H3) and 3 rhesus macaques (M1-M3) were profiled by Quantum Barcoding (QBC; split-pool synthesis) protein measurement and CyTOF mass cytometry, under basal unstimulated conditions. Stimulation-reference analyses use a separate published CyTOF cohort of 46 human and 15 rhesus donors under Basal, CD40L and GM-CSF conditions (Bjornson-Hooper et al 2022. Frontiers in Immunology). MARIO Fusion links cells computationally across assays and individuals, not measurements of the same physical cell. These data support cross-species clustering, MARIO integration, and Bayesian-network inference of conserved causal phospho-signaling relationships. Contents: raw CyTOF .fcs files, raw/processed QBC .fcs and per-cell .csv files, and combined single-cell expression matrices. Version 1.1.0 retains the original source-data archives and adds saved processed fusion assays, matching maps, trajectory fits, network-support tables and numerical results described in manuscript v6. This is a data-and-documentation deposit. Analysis code and processed Seurat objects are provided in the linked separately at GitHub commit repository f0050c36f9f24ccad25b23c52b259c31346df92c.
