Single-cell RNA sequencing (scRNA-seq) has emerged as a powerful tool for investigating cellular heterogeneity, yielding remarkable insights in animal research. However, its application in plant science faces significant challenges due to multiple constraints, including the presence of cell walls, sample processing difficulties, and time sensitivity issues, which have substantially impeded the construction of comprehensive plant cell atlases. While alternative approaches such as single-nucleus RNA sequencing (snRNA-seq) have been proposed, they still exhibit considerable limitations in terms of data quality and reliability. This project aims to overcome the technical bottlenecks in plant scRNA-seq by developing improved protocols specifically designed for recalcitrant plant samples. These advances will significantly enhance the applicability of single-cell transcriptomics in plants, providing crucial technical support for plant developmental biology research and crop improvement efforts.
