This project focuses on the role of vvi-miR395 in grapevine (Vitis vinifera) defense mechanisms against Botrytis cinerea infection. Total RNA samples from grape tissues collected 72 hours post-inoculation were used to construct small RNA and degradome libraries. High-throughput sequencing was performed using an Illumina HiSeq2500 platform, and miRNA sequences were analyzed with ACGT101-miR. Differentially expressed miRNAs were identified using statistical methods (t-test, p < 0.05), and their mRNA targets were predicted through degradome analysis using CleaveLand 4.0 and ACGT101-DEG. Functional annotations of miRNA targets were performed using the GO and KEGG databases to explore their involvement in sulfur metabolism and other defense-related pathways. This study provides new insights into the regulatory role of miRNAs in grapevine immunity and disease resistance.
